Video 1: Sample Preparation
Description:
This video walks you through the Liquid Planktonic Test, the preferred method for sample preparation. You’ll learn how to properly collect, filter, and prepare your sample for ATP testing.

Collecting the Sample: Collect >1 mL of sample in a syringe. The idea volume is 20 to 50 mL.

Filtering the Sample: Attach a filter to the syringe and push at least 1 mL of sample into a waste container. Record the volume filtered.

Handling the Filter (x1): Remove the filter, pull out the plunger, and re-attach the filter. Please handle filter carefully to prevent contamination.

Rinsing the Filter: Filter one bulb-full of Rinse Reagent into a waste container. The rinse reagent to is used remove any organic contaminants from the filter.

Extracting ATP: Pipette 1 mL of Reagent X into the syringe and push it through the filter into a Reagent D tube. Ensure your pipette stays upright at all times.

Storage Options: Close the tube and invert to mix. Your sample is now ready for Video 2: ATP Standard and Sample Reading. ATP extracted in Reagent X remains stable for at least 7 Days. Store the extracted ATP solution in a sealed tube and note any delay when comparing results to ensure accurate interpretation.
Pro Tip: Always wear gloves and avoid breathing on your sample to prevent contamination.
Video 2: ATP Standard and Sample Reading
Description:
This video walks you through the ATP measurement process, including blank readings, standard readings, and sample readings.

Preparing Reagent Z: Prep Reagent Z by adding the liquid buffer to the freeze-dried pellet. Invert to mix. Let it sit for 5 minutes before use.

Turning on your Equipment: Turn on the PhotonMaster by pressing the yellow button. Then press the green button twice to get to the "Ready" screen.

Blank Reading (bRLU): Place an empty assay tube into the chamber. Press on the green button to measure the blank (bRLU) value.

Readings take 10 seconds. Once the reading is complete, record this as your bRLU value.

Standard Reading (sRLU): To measure the standard (sRLU) value, add 100 uL of Reagent Z using the blue pipette and two drops of the Standard to an assay tube.

Tap tube to mix, then insert into the chamber and press the green button to read.

Once the reading is complete, record this as your sRLU value.

Sample RLU: To measure you sample RLU, add 100 uL of Reagent Z to an assay tube using the blue pipette and a new tip.

Next, use the blue pipette and a new tip to add 100 uL of your sample (prepared in Video 1 above).

Tap tube to mix, then insert into the chamber and press the green button to read.

Once the reading is complete, record this as your Sample RLU value.

Calculating your ATP Concentration: Remove your tube from the PhotonMaster once the assay is complete. You are now ready to calculate your ATP concentration.
